It has been found that white blood cells capable of entering the lymph system can serve as a host defense against infection. This defense system is comprised of two portions: the first is the transformation of the macrophages and the second is the migration of the activated lymphocytes.
The first step in the cell-mediated immune response is activation of macrophages. Activation of the macrophages is achieved through the natural production of macrophage stimulating factor (MSF) within activated T cells. Activation of T cells occurs through contact with antigen, thus leading to proliferation and production of MSF in the lymphocyte. An antigen or mitogen is added to the cell culture and causes the lymphocytes to produce macrophage activating factor. After such cell-to-cell contact, the activated macrophages produce a substance termed interferon. Interferon is a substance capable of accelerating white blood cell production in the spleen.
The second step in the cell-mediated immune response is activation of lymphocytes. Activation of the lymphocytes occurs through the interaction of the activated macrophages and the lymphocytes. T lymphocytes provide the second step. The T lymphocytes produce two lymphokines called lymphotoxin and tumuor necrosis factor. These lymphokines cause the lymphocytes to proliferate and migrate. The lymphokines are also responsible for causing the macrophages to lyse and kill intracellular microorganisms.
Accordingly, the activation of the macrophages and the proliferation of the lymphocytes occur in the same mononuclear cell population and are closely related. The proliferation of the T cells and the killing by the macrophages are both affected by the same macrophage activating factor.
In the lymphokines alone, activation of macrophages can be enhanced by interleukin-1. Interleukin-2 is capable of potentiating the macrophage activating effect.
It has been found that by culturing lymphocytes in the presence of macrophage activating factor, lymphokines and lymphocytes can be cultured while awaiting activation. Activation of the macrophages can be accomplished by the addition of suitable
